Gene Description SPOP, speckle-type POZ protein, acts as the substrate targeting component of the cullin-RING-based BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex, to influence degradation of proteins, which includes the BET protein family and therefore, is a putative tumor suppressor (PMID: 19818708, PMID: 28805820, PMID: 30244836, PMID: 28292441). Loss of function Spop mutations in prostate cancer may decrease degradation of BET proteins and confer resistance to BET inhibitors, while gain of function Spop mutations in endometrial cancer may increase degradation of BET proteins and confer sensitivity to BET inhibitors (PMID: 28805821).
